When he was just one year old, his family moved to Happy Canyon Road, Colorado, for a year and then finally settled in Cherry Hills, Arapahoe County, Colorado.

Matt Iseman graduated from Cherry Creek High School in Denver, Colorado, in 1989. Then, he went to Princeton University in Princeton, New Jersey, from where he graduated Cum Laude in 1993 with a Bachelor of Arts degree, majoring in History.
Furthermore, in 1994, he attended Columbia University's Medical School, Physicians, and Surgeons and got his MD in 1998, which was followed by a residency at the University of Colorado Hospital.

After that, Matt connected with his high-school friend, Brad Nieder, and went to a few open mic nights in New York, including the New Talent Night at Comedy Works for a year. Then, he moved to Los Angeles, where he began to perform at comedy clubs, rapidly gaining popularity.
